Abstract: This presentation will describe the iNANO drug delivery model that promotes drug delivery research within an interdisciplinary environment. This talk will focus on our research activities in finding delivery solutions for RNA interference therapeutics through the combined expertise of drug delivery and molecular biology. The ability to utilise synthetic small interfering RNA (siRNA) duplexes to interrupt gene expression is an exciting strategy to silence genes implicated in human disease. Delivery, however, is a key determinant in realising the full clinical potential of RNA interference. Nanoparticle-based technology for overcoming extracellular and intracellular barriers to siRNA (1) will be discussed with focus on a chitosan/siRNA nanoparticle system developed at iNANO (2). We have exploited the predisposition for particle phagocytic capture and mannose-receptor mediated uptake of chitosan as a strategy to treat macrophage-associated inflammatory disease. I will describe our recent work in inflammatory disease models for Rheumatoid Arthritis (RA) (3) and Radiation-Induced Fibrosis (RIF). We demonstrate that knockdown of TNFa expression in systemic macrophages by intraperitoneal (i.p.) administration of chitosan/siRNA nanoparticles in mice reduces systemic inflammation in these two conditions.
